Technical Expertise

I have extensive experience of
  • Molecular biology protocols:

molecular cloning, cDNA sequencing; bacterial cultures; RT-PCR; PCR ; SDM; design, construction & expression of recombinant proteins (fusions and untagged); mRNA isolation; RNA by nuclease protection assays; etc.

  • Protein chemistry:

Extraction & purification of native and recombinant protein from bacteria, baculovirus and animal tissue; low pressure chromatography, FPLC/AKTA; SDS-PAGE & immunoblotting; surface plasmon resonance; ELISA; actin polymerization, fluorescence binding assays, pull-down assays etc.

  • Cellular biology:

maintenance of animal and insect cells in culture; transfection, selection; cell adhesion assays, immunocytochemistry; light, fluorescence & laser confocal microscopy including digital confocal microscopy.

  • Structural biology:

preparation of protein for X-ray crystallography and NMR, seleno-methionine labelling, crystal screening and optimization, derivative screens, data collection, processing and analysis, NMR analysis and interpretation; presentation of structural information thus generated.

  • Computing:

skilled in the use and maintenance of Macintosh® personal computers and software (OS X and earlier). Proficient in UNIX and X11, familiar with IBM-compatibles.

Experience of programming automated systems using Turbo Pascal. Currently getting to grips with C and Objective C. I take pride in the production of universally accessible HTML/CSS. Have used CGI (Perl) and Javascript to advantage in web-based utilities (see http://www2.mrc-lmb.cam.ac.uk/personal/rpg/methods/, specifically the protein and DNA calculators and the codon usage wrangler).

Previous positions
1997 - 1999 (Cambridge Molecular)

Research into and development of manual and automated DNA extraction technologies. I successfully improved existing product lines and maintained their improvement and development. Involved in development of new products. Primary point of liaison between the research and development division and the Kits & Services Product Manager. Administration and re-coding of automated systems software (AP12 & AP96, gNAPS). Project leader of AP12 biology and protocol. Processing requests for material from British Library and checking in of received items. Mac support.
